چکیده

The cerebral cortex is a densely interconnected structure with neural circuits that form between cortical laminae and also between distinct cortical areas. However, the precise cell biological and developmental mechanisms that underlie the formation of these neural circuits remain unknown. Here, we visualize laminar innervation of the developing mouse cerebral cortex by layer II/III pyramidal neurons in real time, describing cytoskeletal dynamics during this process. We find that layer II/III pyramidal neurons achieve local laminar-specific innervation through the stabilization of collateral axon branches in target laminae. We also find that loss of neural activity does not abolish local laminar-specific innervation and that cells within the local environment are the likely source of cues that direct layer-specific cortical innervation.

Fibroblast growth factor 8 regulates neocortical guidance of area-specific thalamic innervation.

Thalamic innervation of each neocortical area is vital to cortical function, but the developmental strategies that guide axons to specific areas remain unclear. We took a new approach to determine the contribution of intracortical cues. The cortical patterning molecule fibroblast growth factor 8 (FGF8) was misexpressed in the cortical primordium to rearrange the area map. Thalamic axons faithfu...

Thalamo-cortical axons regulate the radial dispersion of neocortical GABAergic interneurons

Neocortical GABAergic interneuron migration and thalamo-cortical axon (TCA) pathfinding follow similar trajectories and timing, suggesting they may be interdependent. The mechanisms that regulate the radial dispersion of neocortical interneurons are incompletely understood. Here we report that disruption of TCA innervation, or TCA-derived glutamate, affected the laminar distribution of GABAergi...

Humans and great apes share increased neocortical neuropeptide Y innervation compared to other haplorhine primates

Neuropeptide Y (NPY) plays a role in a variety of basic physiological functions and has also been implicated in regulating cognition, including learning and memory. A decrease in neocortical NPY has been reported for Alzheimer's disease, schizophrenia, bipolar disorder, and depression, potentially contributing to associated cognitive deficits. The goal of the present analysis was to examine var...

Comparative analysis of the nucleus basalis of Meynert among primates.

Long projection axons from the Ch4 cell group of the nucleus basalis of Meynert (nbM) provide cholinergic innervation to the neurons of the cerebral cortex. This cortical cholinergic innervation has been implicated in behavioral and cognitive functions, including learning and memory. Recent evidence revealed differences among primate species in the pattern of cholinergic innervation specific to...

Precise lamination of retinal axons generates multiple parallel input pathways in the tectum.

The axons of retinal ganglion cells (RGCs) form topographic connections in the optic tectum, recreating a two-dimensional map of the visual field in the midbrain. RGC axons are also targeted to specific positions along the laminar axis of the tectum. Understanding the sensory transformations performed by the tectum requires identification of the rules that control the formation of synaptic lami...
